Mapping of genes of innate immunity in domestic breeds of domestic cattle to study the basics of the formation of selection-significant traits

Abstract | The objective of the present work was primary screening for the variability in innate immune genes – Toll-like receptors – in the representative cattle breeds of Russia. The tissue and sperm samples originated from 575 bulls of Kholmogory breed (both of Pechorsky and Severnyi types), Yakut cattle, Yaroslav, Simmental meat-type, Black-Pied and Holstein Black-Pied breeds, as well as buffalo hybrids. Altogether, the samples originated from 17 populations in the collections of of VIZH (All-Russia Institute of Animal Science), VNIIplem (All-Russia Research Institute for Animal Husbandry), Arkhangelsk and Yaroslavl’ central breeding firms. In total, exonic sequences of 10 bovine TLR genes were obtained in seven different cattle breeds. The method of super-long, high-efficiency sequencing allowed for the identification of 64 assumed mutations in coding sequences of the studied genes. In addition, 31 mutations were genotyped using the primer extension technique. The obtained information allows to predict their further occurrence in populations and to use further methods of direct genotyping of animals, e.g. the genotyping microarrays and real-time PCR. |
